What "dual GIP and GLP-1 agonist" means
An agonist is a drug that switches on a receptor, the way a natural hormone would. GLP-1 and GIP are both incretins: hormones your gut releases after you eat that help the pancreas release insulin when blood sugar rises.
- GLP-1 also slows stomach emptying and acts on the brain to reduce appetite and food intake.
- GIP is a second incretin. The Zepbound label says early studies suggest adding GIP may further help regulate food intake. Both receptors are found in brain areas that control appetite.
- Tirzepatide is one molecule that binds and activates both receptors. MedlinePlus groups it with semaglutide as an incretin mimetic.
How much the GIP action adds on its own is still being studied. For head-to-head trial results against semaglutide, see tirzepatide vs semaglutide.
How it compares with other GLP-1 medicines
| Medicine | Receptors it acts on | Status (October 2026) |
|---|---|---|
| Semaglutide (Wegovy, Ozempic, Rybelsus) | GLP-1 | FDA-approved |
| Liraglutide, dulaglutide, exenatide | GLP-1 | FDA-approved |
| Tirzepatide (Zepbound, Mounjaro) | GIP and GLP-1 | FDA-approved |
| Retatrutide | GIP, GLP-1 and glucagon | Investigational; not FDA-approved |
Because tirzepatide already acts on the GLP-1 receptor, the label says not to combine it with any other GLP-1 drug or another tirzepatide product. More on the investigational triple agonist in tirzepatide vs retatrutide.
Is tirzepatide a GLP-2? No
GLP-1 and GLP-2 sound alike, but they are different hormones with different jobs. The GLP-2 medicine, teduglutide, is used for short bowel syndrome in people who need extra nutrition or fluids by IV. It helps the gut absorb fluids and nutrients and isn't a weight-loss drug.
If you see "GLP-2" next to tirzepatide, it's a mix-up. The second hormone tirzepatide targets is GIP.
What this means for you
Tirzepatide shares the same main side effects and warnings as other GLP-1 drugs: nausea and other stomach effects, plus less common risks like pancreatitis and gallbladder disease. See tirzepatide side effects.
Whether tirzepatide or a single-receptor GLP-1 suits you depends on your health history, other medicines, coverage and cost. Through Gabriel, a licensed clinician reviews every prescription request and decides. For prices, see how much tirzepatide costs.
Common questions
Is Mounjaro a GLP-1?
Yes. Mounjaro is tirzepatide, a dual GIP and GLP-1 receptor agonist approved for type 2 diabetes. Zepbound is the same molecule approved for weight and sleep apnea.
Is tirzepatide a GLP-1 or GLP-2?
GLP-1. It activates GLP-1 and GIP receptors. GLP-2 is a different hormone, and the GLP-2 medicine teduglutide is used for short bowel syndrome.
What is the difference between tirzepatide and semaglutide?
Semaglutide acts on the GLP-1 receptor only. Tirzepatide acts on GLP-1 and GIP receptors. Both are weekly injections with similar stomach side effects.
Is tirzepatide a triple agonist?
No. Tirzepatide is a dual agonist. The triple agonist people ask about is retatrutide, which also targets glucagon and is still investigational.
Can you take tirzepatide with another GLP-1?
No. The label says not to combine it with other GLP-1 drugs or other tirzepatide products.
